	<title>Transgenerational Effects of Parental Morphine Withdrawal on Learning and Spatial Memory Processes and Hippocampal BDNF Protein Levels across Two Generations of Male Rats</title>

	<abstract_fa>&lt;strong&gt;مقدمه:&lt;/strong&gt; هدف از این مطالعه، بررسی اثرات ترک مورفین والدینی (مادری و پدری) بر رفتارهای یادگیری و حافظه فضایی و سطوح پروتئین BDNF هیپوکامپ در فرزندان نسل اول و دوم در یک مدل موش صحرایی نژاد ویستار بود. تمرکز پژوهش بر ارزیابی تغییرات رفتاری و شناختی در دو نسل متوالی متعاقب مواجهه والدین با تجویز مورفین و به دنبال آن، دوره ترک است. &lt;strong&gt;مواد و روش&#8204;ها:&lt;/strong&gt; موش&#8204;ها به سه گروه کنترل، گروه ترک مورفین مادری (MMW) و گروه ترک مورفین پدری (PMW) تقسیم شدند. وابستگی به مورفین در والدین با استفاده از دوزهای خوراکی افزایش&#8204;یافته (۱۰ تا ۴۰ میلی&#8204;گرم/کیلوگرم به مدت ۳ هفته) القا شد و سپس یک دوره ترک ۱۰ روزه برای القای فاز ترک مورفین اعمال گردید. پس از تأیید علائم ترک، نسل&#8204;های اول و دوم برای ارزیابی&#8204;های رفتاری پرورش یافتند. رفتار شناختی فرزندان با استفاده از آزمون ماز بارنز ارزیابی شد. پس از آن، موش&#8204;ها کشته شدند و بافت مغز هیپوکامپ برای اندازه&#8204;گیری سطوح پروتئین BDNF هیپوکامپ با استفاده از کیت الایزا استخراج گردید. &lt;strong&gt;یافته&#8204;ها:&lt;/strong&gt; در مرحله یادگیری، زمان&#8204;های تأخیر اولیه در روزهای چهارم و پنجم بین گروه MMW و گروه کنترل تفاوت معنی&#8204;داری نشان داد. تعداد خطاهای اولیه در روز پنجم در نسل اول بین گروه&#8204;های MMW و کنترل تفاوت معنی&#8204;داری داشت. علاوه بر این، تعداد خطاهای اولیه در روز سوم در نسل اول بین گروه&#8204;های PMW و کنترل تفاوت معنی&#8204;داری را نشان داد. در نسل دوم، تعداد خطاهای اولیه نیز تفاوت معنی&#8204;داری را نشان داد، اما تنها در روز سوم بین گروه&#8204;های PMW و کنترل. در آزمون حافظه فضایی (روز ششم)، هر دو گروه MMW و PMW در نسل اول زمان&#8204;های تأخیر و تعداد خطاهای بیشتری را در مقایسه با گروه کنترل نشان دادند. با این حال، در نسل دوم، عوامل زمان تأخیر و تعداد خطاها در مقایسه با گروه کنترل تفاوت معنی&#8204;داری را نشان ندادند. علاوه بر این، سطوح پروتئین BDNF هیپوکامپ در نسل اول در گروه&#8204;های فرزندان دارای والدین با ترک مورفین در مقایسه با گروه کنترل به طور معنی&#8204;داری کاهش یافته بود. &lt;strong&gt;نتیجه&#8204;گیری:&lt;/strong&gt; یافته&#8204;های این مطالعه نشان می&#8204;دهد که ترک مورفین والدینی منجر به اختلالات رفتاری و شناختی معنی&#8204;داری در فرزندان می&#8204;شود. این نتایج بر اهمیت پایش و پیشگیری از مصرف مورفین در والدین به منظور کاهش آسیب&#8204;پذیری در نسل&#8204;های آینده تأکید می&#8204;کند.&lt;br&gt;

	<abstract>&lt;strong&gt;Introduction:&lt;/strong&gt; The aim of this study was to investigate the effects of parental morphine withdrawal (maternal and paternal) on learning and spatial memory behaviors and hippocampal BDNF protein levels in first and second-generation offspring in a Wistar rat model. The focus of the research was on evaluating the behavioral and cognitive changes across two subsequent generations following parental exposure to morphine administration and withdrawal. &lt;strong&gt;Materials and Methods:&lt;/strong&gt; Rats were divided into three groups: control, maternal morphine withdrawal group (MMW), and paternal morphine withdrawal group (PMW). Parental morphine dependence was induced using escalating oral doses (10 to 40 mg/kg for 3 weeks), followed by a 10-day withdrawal period to induce the morphine withdrawal phase. After confirming withdrawal symptoms, first and second generations were bred for behavioral assessments. Cognitive behavior of offspring was evaluated using the Barnes maze test. Afterward, rats were euthanized, and hippocampal brain tissue was extracted to measure hippocampal BDNF protein levels using an ELISA kit. &lt;strong&gt;Results:&lt;/strong&gt; In the learning phase, initial delay times on days four and five showed a significant difference between the MMW group and control. Initial error counts on day five in the first generation differed significantly between the MMW and control groups. Moreover, initial error counts on day three in the first generation showed a significant difference between the PMW and control groups. In the second generation, initial error counts also showed a significant difference, but only on day three between the PMW and control groups. In the spatial memory test (day six), both MMW and PMW groups in the first generation exhibited greater delay times and error counts compared to controls. However, in the second generation, delay time and error count factors did not show a significant difference compared to controls. Furthermore, hippocampal BDNF protein levels in the first generation were significantly reduced in the morphine withdrawal offspring groups compared to the control group. &lt;strong&gt;Conclusion:&lt;/strong&gt; The findings of this study indicate that parental morphine withdrawal results in significant behavioral and cognitive impairments in offspring. These results underscore the importance of monitoring and preventing morphine use in parents to reduce vulnerability in future generations.&lt;span style=&quot;font-size:14.0pt&quot;&gt;&lt;/span&gt;</abstract>
