OTHERS_CITABLE افزایش آستانۀ احساس درد و کاهش بیان ژن گیرندۀ مو-اوپیوئیدی درکبد موش صحرایی با مدل انسفالوپاتی کبدی مقدمه: انسفالوپاتی کبدی یک اختلال مغزی با اختلال عملکردهای شناختی و حرکتی ایجاد شده توسط نارسایی کبد است. هدف از این مطالعه، بررسی تغییرات در آستانه احساس درد و بیان ژن گیرندۀ مو-اوپیوئیدی نوع 1 در کبد موش‌های صحرایی پس از القای انسفالوپاتی کبدی بود. مواد و روش‌ها: موش‌های صحرایی نر (با وزن 280 تا 320 گرم) به طور تصادفی به سه گروه کنترل (بدون عمل جراحی)، شم (با عمل جراحی و بدون بستن مجرای صفراوی مشترک) و گروه با انسداد مجرای صفراوی مشترک، به عنوان یک مدل آزمایشگاهی انسفالوپاتی کبدی، تقسیم شدند. تغییرات در آستانه احساس درد با استفاده از آزمون صفحه داغ، 21 روز پس از انسداد مجرای صفراوی، ارزیابی شد. در روز 21 پس از انسداد مجرای صفراوی، موش‌های صحرایی وزن شده، کشته شدند و کل بافت کبد آن‌ها جدا گردید و وزن شد، سپس درصد وزن کبد به کل بدن محاسبه شد. به علاوه نمونه‌های خون برای بررسی بیوشیمیایی سرم جمع‌آوری شدند و یک قطعه‌ از کبد هر موش صحرایی برای ارزیابی بیان ژن گیرندۀ مو-اوپیوئیدی نوع 1، جداسازی گردید. یافته‌ها: نتایج نشان دادند که آستانه احساس درد، درصد وزن کبد به کل بدن و سطوح سرمی بیلی‌روبین کلی در گروه مدل انسفالوپاتی کبدی در مقایسه با گروه شم به طور معنی‌داری افزایش یافت (0/001>P). نیتروژن اوره خون نیز در این گروه در مقایسه با گروه شم افزایش یافت (0/01>P). همچنین نتایج نشان دادند که بیان ژن گیرندۀ مو-اوپیوئیدی نوع 1در سطوح mRNA در کبد موش‌های صحرایی با انسداد مجرای صفراوی، به طور معنی‌داری در مقایسه با گروه شم کاهش یافت (0/001>P). نتیجه‌گیری: نتایج ما نشان دادند که نارسایی مزمن کبد، بیان ژن گیرندۀ مو-اوپیوئیدی نوع 1 را در کبد تنظیم می‌کند. به علاوه تغییرات در آستانه احساس درد ممکن است یک کندی پردازش حسی را در انسفالوپاتی کبدی تأیید کند. http://shefayekhatam.ir/article-1-643-fa.pdf 2015-05-06 1 10 10.18869/acadpub.shefa.3.1.1 نارسایی کبد بیماری‌های مغز بیلی‌روبین اوره Increase in Nociception Threshold and Decrease in Mu-Opioid Receptor Gene Expression in Liver of a Rat Model of Hepatic Encephalopathy Introduction: Hepatic encephalopathy (HE) is a brain disorder with impairment of cognitive and motor functions caused by liver failure. The aim of this study was to evaluate changes in nociception threshold as well as gene expression of mu-opioid receptor 1 (MOR1) in rat liver after induction of HE. Materials and Methods: The male rats (280-320 g) were randomly divided into three groups of control (non-operated), sham (operated without common bile duct ligation) and a group with a common bile duct ligation (BDL) as an experimental model of HE. Changes in nociception threshold were assessed on day 21 of BDL, using a hotplate test. On day 21 of BDL, rats were weighed, sacrificed and total liver tissue was dissected and weighed, then percentage of liver to body weight was measured. In addition, blood samples were collected for biochemical analysis of serum, and a piece of the liver of each rat was dissected for evaluating the MOR1 gene expression. Results: The results showed that nociception threshold, percentage of liver to body weights, and serum levels of total bilirubin were significantly increased in the HE model group compared to the sham group (P<0.001). Blood urea nitrogen was also increased in this group compared to the sham group (P<0.01). The results also revealed that the MOR1 gene expression at mRNA levels in liver of rats with BDL was significantly decreased compared to the sham group (P<0.001). Conclusion: Our data indicate that chronic liver failure modulates the MOR1 gene expression in liver. OTHERS_CITABLE بررسی تأثیر مهار منتشر شوندۀ قشری مکرر بر میزان بیان گیرنده‌های GABA Aα در هسته تریژمینال مزانسفالیک موش صحرایی نر بالغ مقدمه: مهار منتشر شونده (SD) یک پدیدۀ پاتوفیزیولوژیک است که توسط یک کاهش گذرا در فعالیت نورونی ادامه می‌یابد. مطالعات مختلف نشان می‌دهند که SD می‌تواند در برخی از اختلالات عصبی نقش داشته باشد. هدف مطالعۀ حاضر بررسی اثر SD بر بیان گیرنده‌های GABA A&alpha; در هستۀ تریژمینال مزانسفالیک است. مواد و روش‌ها: حیوانات به طور تصادفی به سه گروه کنترل، شم و تجربی تقسیم شدند. حیوانات بیهوش و در طی جراحی استریوتکس الکترودهای ثبت و کانول تزریق بر روی قشر کاشته شدند. در گروه‌ تجربی محلول کلرید پتاسیم به مدت چهار بار، یکبار در هر هفته، به داخل نئوکورتکس به منظور القای مهار منتشر شوندۀ مکرر تزریق گردید. در گروه شم محلول رینگر تزریق شد و گروه کنترل هیچ مداخله‌ای را دریافت نکرد. در پایان این مرحله حیوانات پرفیوژن و مغزها خارج شدند. پس از آن، بررسی بافت شناسی مغزها و رنگ‌آمیزی ایمونوهیستوشیمی برای بررسی میانگین تعداد نورون‌های تیره و نیز بیان گیرنده‌های GABA A&alpha; و آنزیم GAD65 انجام شد. یافته‌ها: میانگین تعداد نورون‌های تیره در گروه تجربی نسبت به گروه‌های کنترل و شم به طور معنی‌داری افزایش یافت. بیان گیرنده‌های GABA A&alpha; و آنزیم GAD65 در ناحیۀ تریژمینال مزانسفالیک به طور معنی‌داری پس از SD در گروه تجربی نسبت به گروه‌های دیگر کاهش یافت. نتیجه گیری: یافته‌های ما نشان می‌دهند که SD گیرنده‌های مهاری را در ناحیۀ تریژمینال مزانسفالیک مدوله می‌کند. این ممکن است در درمان سردرد مهم باشد. http://shefayekhatam.ir/article-1-650-fa.pdf 2015-05-23 11 20 10.18869/acadpub.shefa.3.1.11 مهار منتشر شوندۀ قشری گاما آمینو بوتیریک اسید نئوکورتکس The Effect of Repetitive Cortical Spreading Depression on the Expression of GABA Aα Receptors in the Mesencephalic Trigeminal Nucleus in Male Adult Rats Introduction: Spreading depression (SD) is a pathophysiologic phenomenon followed by a transient reduction in neuronal activity. Several studies indicate that SD plays a role in some neurological disorders. The present study is aimed to investigate the effect of SD on the expression of GABA A&alpha; receptors in the mesencephalic trigeminal nucleus. Materials and Methods: Animals were randomly classified into three control, sham, and experimental groups. Animals were anesthetized and during the stereotaxic surgery, recording electrodes and the injection cannula were implanted over the cortex. In the experimental group, KCl solution was injected four times once a week into the neocortex to induce repetitive SD. In sham group Ringer solution was injected and control group received no intervention. At the end of this stage, the animals were perfused and the brains were removed. Consequently, histological examination of the brains and immunohistochemical staining method were used to investigate the mean number of dark neurons as well as the expression of GABA A&alpha; receptors and GAD65 enzyme. Results: The mean number of dark neurons significantly increased in the experimental group compared to the sham and control groups. Expression of GABA A&alpha; receptors and GAD65 enzyme in mesencephalic trigeminal region decreased significantly after SD in the experimental group compared to the other groups. Conclusion: Our findings indicate that SD modulates inhibitory receptors in mesencephalic trigeminal region. OTHERS_CITABLE ساب کلونینگ ژن NGF در وکتورترشحی pSecTag2/Hygro و بیان آن در سلول‌های ردۀ PC12 مقدمه: ساب کلونینگ ژن‌های اختصاصی در پلاسمیدها و انتقال آن‌ها به سلول‌های هدف به منظور تولید پروتئین‌های درمانی یا تمایز سلول به عنوان یکی از مؤثرترین روش‌های درمان برای بیماری‌های مختلف پیشنهاد می‌شود. فاکتور رشد عصب (NGF) یکی از اعضای خانواده نوروتروفین‌ها است. نوروتروفین‌ها خانواده‌ای از پروتئین‌ها هستند که میزان بقاء، تمایز و عملکرد انواع مختلف نورون‌ها را تنظیم می‌کنند. مطالعات نشان دادندکه بیان ژن NGF در سلول‌های بنیادی موجب القای تمایز به سمت سلول‌های شبه نورون و رشد آکسون و شاخه‌های آن می‌گردد. مواد و روش‌ها: در این تحقیق هضم آنزیمی بر روی یک پلاسمید حامل ژن NGF انجام گردید و این ژن استخراج شد. ژن NGF در پلاسمید ترشحی pSecTag2 ساب کلون گردید. پلاسمید ساب کلون شده رسوب‌گیری و تغلیظ گردید. سپس با استفاده از لیپوفکتامین به داخل سلول‌های ردۀ PC12 ترانسفکت شد. بیان ژن NGF و تولید پروتئین آن با استفاده از روش‌های RT-PCR و وسترن بلات ارزیابی گردید. یافته‌ها: تعیین توالی نشان داد که فرایند ساب کلونینگ پلاسمید ترشحی درست بوده است. بیان ژن NGF در سلول‌های ترانسفکت شده PC12 به وسیلۀ روش RT-PCR نشان داده شد و تولید پروتئین آن در نتایج وسترن بلات اثبات شد. نتیجه گیری: ساب کلونینگ ژن NGF بر روی وکتور ترشحی pSecTag2 یک روش مناسب جهت انتقال به سلول‌های یوکاریوتی می‌باشد. http://shefayekhatam.ir/article-1-645-fa.pdf 2015-05-06 21 28 10.18869/acadpub.shefa.3.1.21 ترانسفکشن ناقلین ژنتیکی فاکتورهای رشد عصبی سلول‌های PC12 Subcloning of NGF Gene into pSecTag2/Hygro Secretory Vector and Expression in PC12 Cell Line Introduction: Subcloning of specific genes in plasmids and transfecting them into target cells for producing therapeutic proteins or cell differentiation, is suggested as one of the most effective treatment methods for different diseases. Nerve growth factor (NGF) is one of the members of the neurotrophin family. Neurotrophins are a family of proteins that regulate the survival, differentiation, and function of different types of neurons. Studies showed that NGF gene expression in stem cells induces differentiation toward neuron-like cells as well as the growth of axons and its branching. Materials and Methods: In this research, enzymatic digestion on a plasmid carrying NGF gene was performed and extracted. NGF gene was subcloned into pSecTag2/Hygro secretory plasmid. The subcloned plasmid was precipitated and concentrated. It was then transfected by Lipofectamine into PC12 cell line. NGF gene expression and protein production were evaluated using RT-PCR and western blot methods. Results: Sequence determination indicated that secretory plasmid subcloning process has been correct. Expression of NGF gene in transfected PC12 cells was shown by RT-PCR method and production of its protein was proved by the results from western blots. OTHERS_CITABLE بررسی علایم و نشانه‌های عصبی بیماران قبل از دیسککتومی قدامی گردن مقدمه: بیماری‌های تخریب کنندۀ دیسک توسط به هم خوردن ساختارهای آناتومیکی مهره به وجود می‌آیند. زمانی که علایم غیرقابل تحمل هستند یا به درمان‌های غیر جراحی پاسخ نمی‌دهند یا احتمال قوی آسیب به طناب نخاعی وجود داشته باشد، دیسککتومی درمان پیشنهادی است. هدف این مطالعه توضیح علایم بیمار و شدت علایم قبل از جراحی است. مواد و روش‌ها: این مطالعه به صورت گذشته نگر و با مرور پرونده‌های بالینی 103 بیمار انتخاب شده برای دیسککتومی قدامی گردن، انجام شد. داده‌ها توسط آزمون‌های آماری توصیفی آنالیز شدند. یافته‌ها: میانگین سنی بیماران 13/01 ± 50/28 در محدودۀ سنی 25 تا 71 سال بود. میانگین مدت زمان شروع علایم تا بستری شدن برای جراحی 1/94 ± 2/70 ماه بود. 41 نفر از بیماران سیگاری بودند. در تمام موارد، اولین علامت درد گردن بود. رادیکولوپاتی در 55 مورد (53/39%)، رادیکولومیلوپاتی در 44 مورد (42/71%) و میلوپاتی در 4 بیمار (3/88%) وجود داشت. 47 بیمار (45/63%) تخریب دیسک در یک سطح، 53 بیمار (51/45%) در دو سطح و 3 بیمار (2/91%) در سه سطح داشتند. نتیجه گیری: وضعیت بالینی بیماران قبل از دیسککتومی مهم ترین شاخص برای ارجاع سریع بیماران به جراح به شمار می آید. http://shefayekhatam.ir/article-1-652-fa.pdf 2015-05-25 29 35 10.18869/acadpub.shefa.3.1.29 دیسککتومی تخریب دیسک بین مهره‎ای تظاهرات عصبی Investigating the Neurological Signs and Symptoms of Patients before Anterior Cervical Discectomy Introduction: Degenerative disc disease is induced by the collapse of the anatomical structures of the spine. When symptoms are unbearable or does not respond to non-surgical treatments or there is a strong probability to damage to the spinal cord, discectomy is the suggested treatment. The purpose of this study was to explain the patient’s signs and the severity of neurological symptoms prior to surgery. Materials and Methods: This is a retrospective study with review of the clinical records of 103 patients were candidates for anterior cervical discectomy. Data were collected and analyzed by descriptive statistical tests. Results: The mean age of patients was 50.28±13.01 with the range of 25 to 71 years. The mean time from onset of symptoms to hospitalization for surgery was 2.70±1.94 months. Forty-one patients were smokers. In all cases, the first symptom was neck pain. There was radiculopathy in 55 cases (53.39%), radiculomyelopathy in 44 cases (42.71%), and myelopathy in 4 patients (3.88%). Forty-seven patients (45.63%) had disc degeneration at one level, 53 patients (51.45%) at two levels and 3 patients (2.91%) at three levels. OTHERS_CITABLE نقش هسته پشتی- جانبی تالاموس درطول حملات تشنجی غیابی مقدمه: صرع غیابی شاخص، نوعی از صرع ژنرالیزه و غیرتشنجی می‌باشد. ویژگی اصلی این حملات یک اختلال ناگهانی و مختصر در هوشیاری است. اختلالات در حلقۀ تالاموکورتیکال، نقش مهمی را در بیماریزایی تشنج‌های غیابی ایفاء می‌کنند. اگرچه هنوز مشخص نیست که کدام قسمت از این شبکه آغازگر تشنج است. هدف این مطالعه بررسی نقش نورون‌های هستۀ پشتی -جانبی تالاموس در طول تخلیه‌های نیزه و موج‌ها در موش‌های صحرایی نژاد WAG/Rij به عنوان معتبرترین مدل حیوانی صرع غیابی است. مواد و روش‌ها: فعالیت‎های تک واحدی در هستۀ پشتی -جانبی تالاموس و فعالیت الکتریکی مغز از قشر حسی پیکری در موش‌های صحرایی نژاد WAG/Rij شش ماهه در یک زمان ضبط گردید. یافته‌ها: در طول نیزه -موج‌ها، فعالیت‌ تکی در هستۀ پشتی -جانبی تالاموس، تخلیه‌های شبه انفجاری نشان داد که قبل از قله‌های نیزه -موج‌ها شروع شده بودند. در بازه زمانی فارغ از نیزه موج ها، فعالیت‌ شبه انفجاری در هستۀ پشتی -جانبی تالاموس کاهش پیدا کرد. نتیجه گیری: یافته‌های ما پیشنهاد می‌کند که انفجارهای پشت سر هم هستۀ پشتی -جانبی ممکن است قشر را برای آشکار کردن نیزه -موج‌ها تحریک کند. می‌توان اینگونه نتیجه گرفت که مهار انفجار نورون‌های هستۀ پشتی -جانبی شاید به کاهش تکرار نیزه -موج‌ها کمک کند. http://shefayekhatam.ir/article-1-653-fa.pdf 2015-05-25 36 41 10.18869/acadpub.shefa.3.1.36 صرع غیابی تالاموس موش‌های صحرایی قشر حسی -پیکری Role of Laterodorsal Thalamic Nucleus during Absence Seizure Attacks Introduction: A typical absence epilepsy is a type of nonconvulsive and generalized epilepsy. The main feature of these attacks is a sudden brief impairment of consciousness. The disturbances in thalamocortical loop play an important role in pathogenesis of absence seizures. However, it is not clear that which part of this network triggers the seizure. This study was aimed to investigate the role of latrodorsal (LD) thalamic neurons during spike and wave discharges (SWDs) in WAG/Rij rats, as the most valid animal model of absence epilepsy. Materials and Methods: Single unit activities in the LD thalamic nucleus and electrocorticogram of somatosensory cortex were simultaneously recorded in six-month-old WAG/Rij rats. Results: During SWDs, unit activity in the LD thalamic nucleus showed burst-like discharges, which were started before the peak component of SWDs. In SWD-free periods, burst like activity in the LD was reduced. Conclusion: Our findings suggest that the burst firing of LD may stimulate the neocortex to exhibit SWDs. OTHERS_CITABLE تأثیر کاربامازپین برآسیب نورونی در مدل تشنجی پنتیلن تترازول مقدمه: صرع یکی از شایع‌ترین اختلالات سیستم عصبی مرکزی است. مطالعه‌ها نشان دادند که کاربامازپین اثرات ضد صرعی دارد. با این حال هیچ شواهدی مبنی بر اینکه کاربامازپین اثرات محافظتی بر آسیب نورونی القاء شده توسط تشنج دارد، وجود ندارد. در این مطالعه اثرات کاربامازپین بر روی آسیب سلولی مشاهده شده در مدل تشنج پنتیلن تترازول (PTZ) در مغز موش صحرایی مورد ارزیابی قرار گرفت. مواد و روش‌ها: رفتار تشنجی و ارزیابی بافت‌شناسی در موش‌های صحرایی با وزن و سن برابر انجام شد، که به 5 گروه تقسیم شدند: 1. گروه کنترل: بدون هیچ گونه مداخله، 2. گروه PTZ: پس از تزریق PTZم(40 میلی گرم/کیلوگرم)، 3. گروه شم: حلال‌ها 30 دقیقه قبل از تزریق PTZم(40 میلی گرم/کیلوگرم) تزریق شدند، 4. گروه درمان 1: کاربامازپین (10 میلی گرم/کیلوگرم) 30 دقیقه قبل از تزریق PTZ تزریق شد، 5. گروه درمان 2: کاربامازپین (40 میلی گرم/کیلوگرم) 30 دقیقه قبل از تزریق PTZ تزریق شد. یافته‌ها: کاربامازپین به طور معنی‌داری تولید نورون‌های تیره را در نواحی CA1 و CA3 هیپوکامپ و قشر گیجگاهی پس از تشنج القاء شده توسط PTZ در مقایسه با گروه‌های PTZ و شم کاهش داد. تراکم نورون‌های تیره در گروه‌های درمانی به میزان معنی‌داری از گروه کنترل بیشتر بود. نتیجه گیری: نتایج نشان دادند که کاربامازپین به عنوان ماده حفاظت‌کننده نورونی پس از حمله‌های تشنجی عمل می‌نماید. http://shefayekhatam.ir/article-1-654-fa.pdf 2015-05-25 42 48 10.18869/acadpub.shefa.3.1.42 کاربامازپین صرع تشنج پنتیلن تترازول هیپوکامپ The Effect of Carbamazepine on Neuronal Damage in Pentylentetrazole Model of Seizure Introduction: Epilepsy is the most common disorder of the central nerves system. Studies showed that carbamazepine has anti-epileptic effects. However, there is no evidence whether carbamazepine protect seizure-induced neuronal injury. In this study, the effects of carbamazepine were evaluated on cell injury observed in pentylenetetrazole (PTZ) model of seizure in the rat brain. Materials and Methods: The convulsive behaviour and histological evaluation were performed in age- and weight-matched rats, which were divided into five groups: 1.control group: without any intervention, 2. PTZ group: after PTZ injection (40 mg/ kg), 3. Sham group: vehicles were injected 30 min before PTZ injection (40 mg/ kg), 4. Treatment group 1: carbamazepine (10 mg/kg) was injected 30 min before PTZ injection, 5. Treatment group 2: carbamazepine (40 mg/kg) was injected 30 min before PTZ injection. Results: Carbamazepine significantly decreased production of the dark neurons in the hippocampal CA1 and CA3 areas and the temporal cortex after PTZ-induced seizure compared to PTZ and sham groups. Density of dark neurons in treatment groups was significantly higher than control group. OTHERS_CITABLE اثر عصارۀ مغز آسیب دیده بر روی تکثیر سلول‎های بنیادی عصبی کشت شده در محیط سه بعدی مقدمه: پس از آسیب اولیۀ بافت مغز، مکانیسم‎های ثانویه‎ای فعال می‎شوند و منجر به آزادسازی فاکتورهای حمایتی و نوروتروفیک مختلف در بافت‌های آسیب دیده می‌گردند. تأثیر این محیط جدید بر نورون‎زایی، تکثیر و بقاء سلول‎های بنیادی نیاز به بررسی‌های بیشتری دارد. هدف از مطالعۀ حاضر بررسی تأثیر عصارۀ مغز آسیب دیده بر تکثیر سلول‎های بنیادی عصبی جنینی موش صحرایی بود. مواد و روش‎ها: سلول‎های بنیادی عصبی از برجستگی‎های عقده‎ای جنین موش صحرایی جدا شدند، سپس به عنوان نوروسفر کشت داده شدند. در مرحله بعد، سلول‎ها در داربست پوراماتریکس به عنوان کشت سه بعدی، کشت داده شدند. بر اساس محتوای محیط کشت، سلول‌ها به 4 گروه: با فاکتور رشد، فاقد فاکتور رشد، فاقد فاکتور رشد + عصارۀ مغز سالم و فاقد فاکتور رشد + عصارۀ مغز آسیب دیده، تقسیم شدند. ارزیابی تکثیر توسط مشاهدۀ سلول‎های مشخص شده با DiI انجام شد و سنجش بقاء توسط آزمایش MTS پس از 10 روز انجام گردید. جهت آماده سازی عصاره مغز آسیب دیده، از یک مدل آسیب مغزی موش صحرایی استفاده گردید و عصاره 48 ساعت پس از آسیب مغزی جمع آوری شد. یافته‌ها: نتایج نشان دادند که سلول‎های بنیادی عصبی مشتق شده از برجستگی‎های عقده‎ای جنینی موش صحرایی، توانایی تکثیر بالایی دارند. سلول‎های DiI مثبت و آزمایش MTS تمایل بالاتری را به تکثیر و بقاء سلول‎های بنیادی عصبی در گروه‌‌های فاقد فاکتور رشد + عصارۀ مغز آسیب دیده و حاوی فاکتور رشد نسبت به گروه‌های فاقد فاکتور رشد و فاقد فاکتور رشد + عصارۀ مغز سالم، نشان دادند. نتیجه گیری: نتایج ما یک اثر مثبت احتمالی عصارۀ مغز آسیب دیده را بر روی بقاء و تکثیر سلول‏های بنیادی عصبی جنینی موش صحرایی نشان داد. مطالعات بیشتر به منظور بررسی جزئیات یافته‌های اولیه ما مورد نیاز است. http://shefayekhatam.ir/article-1-646-fa.pdf 2015-05-07 49 56 10.18869/acadpub.shefa.3.1.49 سلول‌های بنیادی عصبی آسیب‌های مغزی عصاره‌های بافتی پوراماتریکس Effect of Injured Brain Extract on Proliferation of Neural Stem Cells Cultured in 3-Dimensional Environment Introduction: After primary brain injury, secondary mechanisms are activated and lead to releasing of various supportive and neurotrophic factors in injured tissues. The effect of the new environment on neurogenesis, proliferation, and survival of stem cells needs more investigations. The aim of the present study was to evaluate the effect of injured brain extract on proliferation of embryonic rat neural stem cells (NSCs). Materials and Methods: NSCs were isolated from ganglionic eminences of embryonic rat then cultured as neurospheres. Next, cells were seeded in PuraMatrix scaffold as 3-dimension culture. Based on the medium content, cells were divided into 4 groups: with growth factor, without growth factor, without growth factor + intact brain extract, and without growth factor + injured brain extract. Proliferation assay was done by evaluation of the DiI labeled cells and the survival assay was carried out by MTS test 10 days later. For preparation of the injured brain extract, a rat brain injury model was utilized and the extract was collected 48 hours after brain injury. Results: The results showed that NSCs derived ganglionic eminence of embryonic rat had high proliferation ability. DiI-positive cells and MTS test showed a higher tendency of the proliferation and survival of NSCs in the without growth factor + injured brain extract and growth factor groups compared to the without growth factor and without growth factor + intact brain extract groups. Conclusion: Our results indicated a possible positive impact of injured brain extract on survival and proliferation of rat embryonic neural stem cells. OTHERS_CITABLE نقش گیرنده AMPA در هسته قاعده‌ای -جانبی آمیگدال بر روی پرخاشگری و اضطراب در موش‌های صحرایی مقدمه: براساس مطالعه‌های قبلی مسیر نورونی هستۀ قاعده‌ای -جانبی آمیگدال یک نقش مهمی را در سبب شناسی و پاتوفیزیولوژی افسردگی، اضطراب و خشونت بازی می‌کند. با این حال نقش دقیق هستۀ قاعده‌ای -جانبی آمیگدال در اضطراب نیاز به توضیح دارد. با استفاده از ارتباط میان‌کنش اجتماعی و آزمایش‌های ماز بعلاوه‌ای شکل مرتفع، نقش زیرواحدهای گیرنده گلوتامات AMPA در هستۀ قاعده‌ای -جانبی آمیگدال روی پرخاشگری و رفتارهای شبه اضطرابی بررسی شد. مواد و روش‌ها: پس از بیهوشی، دو کانول در هستۀ قاعده‌ای -جانبی آمیگدال در موش‌های صحرایی ویستار جای داده شدند. پس از یک هفته بهبودی، سطوح اضطراب و اشکال گوناگون پرخاشگری توسط آزمون‌های ماز بعلاوه‌ای شکل مرتفع و میان‌کنش اجتماعی بررسی شدند. یافته‌ها: آگونیست AMPA در دوزهای 0/25، 0/5 و 1 میلی‌گرم/کیلوگرم در هسته قاعده‌ای -جانبی آمیگدال تجویز شد. دوزهای 0/5 و 1 میلی گرم/کیلوگرم به طور معنی‌داری زمان سپری شده در بازوهای باز و پرخاشگری از طرفین را در مقایسه با گروه شم افزایش دادند. علاوه بر این، فعالسازی گیرندۀ AMPA در دوز 1 میلی‌گرم/کیلوگرم درصد ورود و ماندن در بازوی باز و پرخاشگری از طرفین را به طور معنی‌داری کاهش می‌دهد. نتیجه گیری: نتایج ما نشان می‌دهند که گیرنده‌های AMPA پیام‌رسانی در هستۀ قاعده‌ای -جانبی را تعدیل می‌کنند و ممکن است اضطراب و پرخاشگری را تحت تأثیر قرار دهند. http://shefayekhatam.ir/article-1-655-fa.pdf 2015-05-27 57 64 10.18869/acadpub.shefa.3.1.57 گیرنده‌های AMPA آمیگدال اضطراب پرخاشگری The Role of AMPA Receptor in the Basolatral Amygdala on Aggression and Anxiety in Rats Introduction: According to the previous studies, neural pathway of the basolatral amygdala plays an important role in etiology and pathophysiology of depression, anxiety, and aggression. However, the exact role of the basolateral amygdala in anxiety needs to be elucidated. Using social interaction and elevated plus maze tests, the role of AMPA glutamate subreceptors in the basolatral amygdala on aggression and anxiety-like behaviors was investigated. Materials and Methods: After anesthesia, two cannulas were implanted in the basolatral amygdala in Wistar rats. After one week recovery, anxiety levels and different forms of aggression were analyzed by elevated plus maze and social interaction tests. Results: AMPA agonist was administered at 0.25, 0.5, and 1 mg/kg doses in the basolateral amygdala. Doses of 0.5 and 1 mg/kg were significantly increased time spent in the open arms and offensive sideway compared to sham group. In addition, activation of AMPA receptor at 1 mg/kg significantly decreased the stay and entrance percentage of open arm and offensive sideway. OTHERS_CITABLE شیوع اضافه وزن و چاقی در بیماران آسیب نخاعی مقدمه: چاقی یکی از شایع‌ترین مشکلات مرتبط با رژیم غذایی می‌باشد و خطر بیماری‌های دیابت شیرین، فشارخون بالا، تصلب شرایین و چربی خون بالا را افزایش می‌دهد. بیماری‌های قلبی -عروقی یکی از علل اصلی ناخوشی و مرگ و میر در افراد مبتلا به آسیب نخاعی هستند. افزایش تودۀ چربی به عنوان یک عامل خطر مهم در بیماران با آسیب نخاعی مزمن مشخص شده است و کنترل وزن به عنوان یک استراتژی مهم در پیشگیری از بیماری‌های قلبی -عروقی توصیه می‌شود. هدف از این مطالعه، بررسی میزان شیوع چاقی و اضافه وزن در افراد دارای آسیب نخاعی می‌باشد. مواد و روش‏ها: یک مطالعه به صورت مشاهده‌ای مقطعی -تحلیلی بر روی پرونده‌های 850 بیمار مبتلا به آسیب نخاعی مراجعه کننده به بیمارستان خاتم الانبیای تهران، ایران از سال‌های 1387 تا 1392 انجام شد. این اطلاعات شامل سن، جنس، قد، وزن، مدت زمان گذشته از آسیب نخاعی و سطح آسیب نخاعی می‌باشد. سپس نمایۀ تودۀ بدنی برای هر بیمار محاسبه شد و شیوع اضافه وزن و چاقی تعیین شد. یافته‏ها: از 850 بیمار مورد مطالعه 833 نفر مرد و 17 نفر زن بودند. در کل، 50/2% بیماران نمایۀ تودۀ بدنی طبیعی داشتند و 7/6% از بیماران دچار سوءتغذیه بودند. شیوع اضافه وزن و چاقی به ترتیب 32/1% و 10% بود. یک رابطۀ معنی‌داری بین سن و نمایۀ تودۀ بدنی وجود داشت اما هیچ همبستگی معنی‌داری بین سطح آسیب و نمایۀ تودۀ بدنی یافت نشد. نتیجه گیری: شیوع اضافه وزن و چاقی در افراد دارای آسیب نخاعی بالا است و یک همبستگی مثبت معنی‌داری بین سن و نمایۀ تودۀ بدنی وجود دارد. http://shefayekhatam.ir/article-1-656-fa.pdf 2015-05-27 65 70 10.18869/acadpub.shefa.3.1.65 آسیب‌های طناب نخاعی چاقی نمایۀ تودۀ بدنی Prevalence of Overweight and Obesity in Patients with Spinal Cord Injury Introduction: Obesity is one of the most prevalent diet-related problems and increases the risk for diabetes mellitus, hypertension, atherosclerosis, and dyslipidemia. Cardiovascular diseases are one of the major of morbidity and mortality in people with a spinal cord injury (SCI). Increased fat mass has also been identified as an important risk factor in chronic SCI and weight management is recommended as an important preventive strategy of cardiovascular disease. The aim of this study is to determine the prevalence of overweight and obesity in people with SCI. Materials and Methods: An observational analytic cross sectional study was done on 850 SCI patients registered in Khatam Alanbia Hospital, Tehran, Iran, from 2008 to 2013. These data are included age, sex, height, weight, duration of SCI, and the level of SCI. The body mass index (BMI) was subsequently calculated for each patient and the prevalence of overweight and obesity were determined. Results: Of the 850 patients, 833 patients were male and 17 were female. In all, 50.2% of patients had a normal BMI and 7.6% of patients were undernourished. The prevalence of overweight and obesity were 32.1% and 10%, respectively. There was a significant relation between age and body mass index but there was no significant correlation between the level of injury and body mass index. OTHERS_CITABLE بررسی اثر عصارۀ ریشه گزنه بر پارامترهای هیستومتریک قشر مخ و مخچه در موش‌های صحرایی متعاقب تجویز تستوسترون مقدمه: تستوسترون یک هورمون استروئیدی است که توسط گنادها و غدد آدرنال ترشح می‌شود. دوز بالای تستوسترون منجر به کاهش نورون‌ زایی می‌شود و آپوپتوز را در شرایط آزمایشگاهی القاء می‌کند. جهت تعدیل اثر تستوسترون، گیاهان حاوی ویژگی‌های آنتی آندروژن می‌توانند مفید باشند. اورتیکا دیوئیکا، که اغلب به نام گزنه رایج است، حاوی ترکیبات آنتی آندروژنی می‎باشد. علاوه بر این، اثرات فیتواستروژنی گزنه ارزیابی شده است. در این مطالعه اثر عصارۀ ریشۀ گزنه بر ساختارهای بافتی قشر مخ و مخچۀ موش‌های صحرایی بعد از تزریق تستوسترون مورد ارزیابی قرار گرفت. مواد و روش‎ها: 20 سر موش صحرایی نژاد ویستار بالغ نر سالم به صورت تصادفی به 4 گروه تقسیم شدند: گروه کنترل (تغذیۀ معمولی بدون هیچ‌گونه درمانی دریافت کردند)، گروه تستوسترون (10 میلی‌گرم/کیلوگرم تستوسترون به صورت زیر جلدی دریافت کردند)، گروه عصارۀ ریشۀ گزنه (50 میلی‌گرم/کیلوگرم عصارۀ ریشۀ گزنه به صورت خوراکی دریافت کردند) و گروه گزنه + تستوسترون (50 میلی‌گرم/کیلوگرم عصارۀ ریشۀ گزنه به صورت خوراکی و 10 میلی‌گرم/کیلوگرم تستوسترون دریافت کردند). پس از 6 هفته، مغزها توسط هماتوکسیلین و ائوزین رنگ آمیزی شدند. تعداد جسم سلولی نورونی، قطر هستۀ نورون‌ها، قطر سلول‌های پورکنژ و قطر لایه‌های مخچه اندازه گیری شد. یافته‌ها: هیچ تفاوت معنی‌داری در میانگین تعداد جسم سلولی نورونی در قشر مخ، قطر سلول‌های پورکنژ در مخچه و ضخامت لایه‌های مخچه بین گروه‌های مختلف وجود نداشت. قطر هستۀ نورون‌ها در لایۀ گرانولار داخلی مخ در گروه تستوسترون نسبت به گروه‌های گزنه و گزنه + تستوسترون به طور معنی داری افزایش یافت. نتیجه گیری: گیاه گزنه می‌تواند به عنوان یک تعدیل کنندۀ تستوسترون در نظر گرفته شود. جهت تعیین اثر دقیق‏تر گزنه بر مغز، مطالعه‌های بیشتری مورد نیاز است. http://shefayekhatam.ir/article-1-659-fa.pdf 2015-05-30 71 78 10.18869/acadpub.shefa.3.1.71 مغز موش‏های صحرایی اورتیکا دیوئیکا تستوسترون Effect of Nettle Root Extract on Histometrical Parameters of Cerebral and Cerebellar Cortices in Rat Following Administration of Testosterone Introduction: Testosterone is a steroid hormone secreted by gonads and adrenal glands. High dose of testosterone leads to decrease neurogenesis and induce apoptosis in vitro. To modulate testosterone effect, plants contain anti-androgenic properties can be useful. Urtica dioica, often called common nettle, contains anti-androgen compounds. Furthermore, the phytoestrogens effects of nettle have been evaluated. In this study, the effect of nettle root extract on histological structures of the cerebral and cerebellar cortices of rats was evaluated after testosterone injection. Materials and Methods: Twenty healthy adult male Wistar rats were divided randomly into 4 groups: control group (received ordinary feed without any treatment), testosterone group (received 10 mg/kg testosterone subcutaneously), nettle root extract group (received 50 mg/kg nettle root extract orally), nettle + testosterone group (received 50 mg/kg orally nettle root extract and 10 mg/kg testosterone). After 6 weeks, the brains were stained by hematoxylin and eosin. The number of neuronal cell body, nuclear diameter of neurons, diameter of Purkinje cells, and diameter of cerebellar layers were measured. Results: There were no significant differences in the mean number of neuronal cell body in cerebral cortex, diameter of Purkinje cells in cerebellum, and thickness of cerebellar layers among different groups. The nuclear diameter of neurons in inner granular layer of cerebrum in the testosterone group significantly increased compared to the nettle and nettle + testosterone groups. Conclusion: Nettle plant can be considered as a testosterone modulator. CASE_STUDY خونریزی زیر سخت شامه‎ای شبیه خونریزی اپیدورال: یک گزارش مورد مقدمه: آسیب‌های مغزی ناشی از ضربه، آسیب‌های پیچیده با یک محدوده وسیعی از علایم و ناتوانی‌ها هستند. آسیب‌های مغزی ناشی از ضربه شامل خونریزی اپیدورال، خونریزی زیر سخت شامه‎ای، خونریزی داخل مغزی، خونریزی زیر عنکبوتیه، آسیب منتشر آکسونی و کوفتگی مغزی می‌باشد. توصیف بیمار: بیمار یک مرد 22 ساله بود که به اورژانس بیمارستان امام خمینی (ره) ارومیه، ایران با شکایت سردرد مراجعه کرد. شدت سردرد حدود 8 از 10 (بر اساس مقیاس آنالوگ بصری) بود. بیمار به طور کامل هوشیار بود و مقیاس کومای گلاسکو 15 بود. سی تی اسکن مغز، خونریزی اپیدورال و وجود خون را نشان داد که شیار سیلوین را پر کرده بود. یافته‌ها: در طی جراحی آشکار شد که بیمار از خونریزی زیر سخت شامه‎ای همراه با خونریزی در یک کیست عنکبوتیه مزمن رنج می‌برد. نتیجه گیری: خونریزی زیر سخت شامه‎ای تجمع خون در فضای بین سخت‌شامه و لایه زیرعنکبوتیه است و از وریدها منشأ می‌گیرد. خونریزی زیر سخت شامه‎ای در افراد الکلی و بیماران مسن تر، شایع ‌تر است. در موارد نادری خون در کیست عنکبوتیه جمع می‌شود و شبیه خونریزی زیر عنکبوتیه دیده می‌شود. http://shefayekhatam.ir/article-1-644-fa.pdf 2015-05-06 79 83 10.18869/acadpub.shefa.3.1.79 خونریزی زیر سخت شامه‌ای خونریزی جمجمه‌ای اپیدورال مغز خونریزی زیر عنکبوتیه Subdural Hematoma Mimics Epidural Hematoma: A Case Report Introduction: Traumatic brain injuries (TBI) are complex injuries with a broad range of symptoms and disabilities. TBI include epidural hematoma (EDH), subdural hematoma (SDH), intracerebral hemorrhage, subarachnoid hemorrhage (SAH), diffuse axonal injury, and brain contusion. Case Description: The patient was a 22-year-old man referred to the emergency department of Imam Khomeini hospital, Urmia, Iran, suffering from headache. Headache severity was 8 out of 10 (according to visual analog scale). The patient was fully alert and Glasgow coma scale was 15. Brain CT scan showed EDH and blood that filled the sylvain fissure. Results: It was revealed during surgery that patient was suffering from SDH with bleeding in a chronic arachnoid cyst. Conclusion: SDH is the collection of blood in the space between the dura and subarachnoid layer and originating from veins. SDH is more common in alcoholic and older patients. REVIEW_ARTICLE کاربرد سلول‌های بنیادی در ترمیم صدمه‌های اعصاب محیطی: یک مطالعه مروری مقدمه: سیستم اعصاب محیطی از قابلیت ترمیم ذاتی برخوردار است که بسیاری از عوامل مانند سلول‌های شوآن، فاکتورهای رشد و ماتریکس خارج سلولی در شکل‌گیری آن درگیر هستند. هرچند در صدمه‌های شدید، فرایند ترمیم اعصاب محیطی بسیار ضعیف و ناکارآمد است و اقدام‌های درمانی جهت بازسازی عصب مورد نیاز هستند. روش‌های درمانی حاضر محدودیت‏های متعدد و کارایی پایینی دارند. در این راستا، یکی از مهم ترین رویکردهای پژوهشی به کارگیری سلول‏های بنیادی در ترمیم اعصاب محیطی است. یکی از مزایای این سلول‌ها، توانایی آن‌ها برای تمایز به سلول‌های شوآن می‌باشد. سلول‌های بنیادی عملکرد سیستم ایمنی را توسط ترشح فاکتورهای نوروتروفیک تعدیل می‌کنند و به تشکیل لایۀ میلین در طی ترمیم اعصاب محیطی کمک می‌کنند. تحقیقات در این زمینه نیز این قابلیت‏ها را نشان می‌دهند و آینده روشنی را در کاربرد سلول‌های بنیادی در ترمیم جراحت‌های اعصاب محیطی نوید می‎دهند. نتیجه گیری: با توجه به اهمیت سلول‏های بنیادی در آیندۀ پزشکی ترمیمی و مهندسی بافت عصبی، درک ویژگی‎های سلول‎های بنیادی و نیز شناخت منابع استخراج و قابلیت‌های آن‌ها در پیشبرد ترمیم اعصاب محیطی ضروری می‌باشند. این مقاله مروری است بر مهم‌ترین پیشرفت‌هایی که در این زمینه به دست آمده است. http://shefayekhatam.ir/article-1-660-fa.pdf 2015-05-31 84 98 10.18869/acadpub.shefa.3.1.84 اعصاب محیطی ترمیم سلول‌های شوآن سلول‌های بنیادی Use of Stem Cells in the Regeneration of Peripheral Nerve Injuries: an Overview Introduction: Peripheral nervous system has an innate regenerative ability that many factors are involved in its formation, such as Schwann cells, growth factors, and extracellular matrix. However, in severe injuries, peripheral nerve regeneration process is very weak and ineffective and therapeutic measures are needed for nerve regeneration. Current therapeutic methods have several limitations and low efficacy. In this regard, one of the most important research approaches is using stem cells in the regeneration of peripheral nerves. Stem cells have a potential to differentiate into Schwann cells. Stem cells modulate the immune system by secreting neurotrophic factors and help formation of myelin layer during peripheral nerve regeneration. Researches in this field also represent these capabilities and promise a bright future in the application of stem cells in the regeneration of peripheral nerve injuries. Conclusion: Regarding the importance of stem cells in the future of regenerative medicine and neurological tissue engineering, understanding of characteristics of stem cells as well as recognition of the extraction resource and their abilities in promoting the peripheral nerve regeneration are necessary. A vast amount of data indicates the potential of stem cell therapy for various neurological diseases. In the present review, the possible beneficial effects of stem cells for treatment of nervous system disorders were presented. The stem cells have been suggested for treatment of different acute and chronic nervous system disorders such as hemorrhagic as well as ischemic stroke, Parkinson's disease, Hantington's disease, amyotrophic lateral sclerosis, multiple sclerosis, and Alzheimer's disease. REVIEW_ARTICLE پیش‌ آماده‌سازی با فعالیت ورزشی و حفاظت عصبی: مروری بر مکانیسم‏ها مقدمه: حوادث عروق مغزی یا سکتۀ مغزی نخستین عامل ناتوانی و سومین علت مرگ و میر در افراد بزرگسال به شمار می‌رود. مشخص شده است که ممکن است فعالیت ورزشی با تعدیل عوامل خطری مانند فشارخون بالا، پروفایل لیپیدی و دیابت، نقش مهمی را در پیشگیری از حوادث عروق مغزی ایفاء کند. در هر حال یافته‌های اخیر پیشنهاد می‏کنند که فعالیت ورزشی علاوه بر تعدیل عوامل خطر سکته مغزی، ممکن است در القای حفاظت عصبی اندوژن و حفظ حیات نورون‏ها در شرایط آسیب ایسکمی -خونرسانی مجدد، که مکانیسم اصلی سکتۀ مغزی ایسکمیک می‌باشد، تأثیر داشته باشد. تأثیر فعالیت‏های ورزشی پیشین بر حفاظت نورونی در مقابل آسیب ایسکمی و القای مقاومت نورونی به عنوان پیش آماده سازی فعالیت ورزشی نامیده می‌شود، که حیطۀ مطالعات پژوهشی جدیدی در زمینۀ تأثیرات فعالیت ورزشی بر مغز به شمار می‌رود. اگرچه مکانیسم دقیق حفاظت عصبی ناشی از پیش آماده ‌سازی با فعالیت ورزشی هنوز به طور کامل مشخص نشده است، مطالعات گذشته نشان داده‌اند که پیش آماده ‌سازی با فعالیت ورزشی ممکن است از طریق چندین مکانیسم‏ از قبیل تقویت سد خونی -مغزی، القای عروق زایی و ایجاد شریان‌های جدید مغزی، بهبود متابولیسم مغز و کاهش اختلالات متابولیکی مغز متعاقب آسیب ایسکمی، تنظیم افزایشی بیان نوروتروفین‎ها و نیز کاهش التهاب، آپوپتوز و استرس اکسیداتیو به القای حفاظت عصبی در مغز کمک کند. نتیجه گیری: به نظر می‏رسد که ممکن است پیش آماده سازی با فعالیت ورزشی به کاهش آسیب اولیه و بهبود نتایج نورولوژیک بعد از آسیب ایسکمی -خونرسانی مجدد، در افراد مستعد به آسیب‌های ایسکمیک مغزی یا در افراد با سابقۀ آسیب ایسکمیک خفیف، منجر شود. با این وجود مطالعات بیشتر برای بهبود راهکارهای فعالیت‌های ورزشی با مدت، شدت و نوع مناسب جهت القای حفاظت عصبی بهینه مورد نیاز است. http://shefayekhatam.ir/article-1-648-fa.pdf 2015-05-09 115 130 10.18869/acadpub.shefa.3.1.115 فعالیت ورزشی سکته مغزی ایسکمی Exercise Preconditioning and Neuroprotection: A Review of Mechanisms Introduction: Cerebrovascular accident or stroke is the first cause of acquired disability and the third leading cause of mortality in adults. It is known that exercise modifies risk factors such as hypertension, lipid profile, diabetes that may play an important role in the prevention of cerebrovascular accidents. However, recent findings suggest that besides adjusting the risk factors of stroke, exercise may be helpful in inducing endogenous neuroprotection and neuronal survival in ischemia – reperfusion condition which is the main mechanism of ischemic stroke. The effect of previous exercises in protecting neurons against ischemic injury and inducing neuronal resistance known as exercise preconditioning, which is a relatively new field of research on the effects of exercise on the brain. Although the exact mechanisms of neuroprotection induced by exercise preconditioning have yet to be known, previous studies have shown that exercise preconditioning may be helpful through several mechanisms, such as strengthening blood-brain barrier, inducing cerebral angiogenesis and arteriogenesis, improving cerebral metabolism and decreasing neuronal metabolic disturbances following ischemic injury, upregulation of neurotrophins expression, as well as reducing inflammation, apoptosis, and oxidative stress. Conclusion: It seems that exercise preconditioning in people predisposed to brain ischemic injuries or in people with history of mild ischemic injury may help in reducing the primary damage and improve the neurological outcomes after ischemia-reperfusion injury. REVIEW_ARTICLE نقش میکرو RNA ها در تکوین سلول‌های ایمنی و سیستم عصبی و ارتباط آن‌ها با مالتیپل اسکلروز مقدمه: میکرو RNA ها ریبونوکلئیک اسیدهای کوچک و غیر کدکننده‌ هستند که در تنظیم بیان ژنوم میزبان در سطح پس از رونویسی نقش‌های اساسی ایفاء می‌کنند. یک میکرو RNA به تنهایی می‌تواند موجب تنظیم کاهشی چندین رونوشت mRNA ی هدف شود. بنابراین تغییرات اندک در بیان یک میکرو RNA ممکن است منجر به تغییرات چشمگیری در بیان ژن‌های مختلفی ‌شود. در طی دو دهۀ اخیر میکرو RNA ها به عنوان تنظیم‌کننده‌های کلیدی تمایز دودمان سلول ایمنی، بلوغ، حفظ هومئوستاز و عملکرد طبیعی ایمنی پدیدار شده‌اند. مالتیپل اسکلروز یک بیماری التهابی مزمن است که با نفوذ لنفوسیت‌ها به سیستم عصبی مرکزی، از دست رفتن میلین و تخریب آکسونی مشخص می شود. با وجود اینکه علل مالتیپل اسکلروز تاکنون ناشناخته مانده است، به‌طور گسترده‌ای پذیرفته شده است که اهداف دارویی جدید به منظور تمرکز بر روی کاهش التهاب و تقویت فرایند ترمیم سیستم عصبی مرکزی لازم است. تحقیقات اخیر درباره بیماری مالتیپل اسکلروز نشان داده‌اند که میکرو RNA ها در سیستم ایمنی و سیستم عصبی مرکزی دچار اختلال می‌شوند، که نقش آن‌ها را در روند بیماریزایی مالتیپل اسکلروز نشان می‌دهد. نتیجه‌گیری: شناسایی الگوهای خاص بیان میکرو RNA در بیماری‌های خودایمنی و یک فهم جامع از نقش آن‌ها در روند بیماریزایی بیماری‌های مختلف نه تنها شناساگرهای تشخیصی مولکولی جدید، بلکه همچنین استراتژی‌های ژن‌درمانی جدیدی را برای درمان بیماری‌های خودایمنی التهابی پیشنهاد می‌کند. در این مطالعه ما آخرین یافته‌های مربوط به تولید و تکامل میکرو RNA و اثر آن در سیستم عصبی مرکزی و سلول‌های ایمنی بیماران مالتیپل اسکلروز را مرور می‌کنیم. http://shefayekhatam.ir/article-1-647-fa.pdf 2015-05-07 131 144 10.18869/acadpub.shefa.3.1.131 میکرو RNA ها التهاب خود ایمنی سیستم عصبی مرکزی بیماری‌های از بین برندۀ میلین Role of MicroRNAs in Development of Immune Cells and Nervous System and their Relation to Multiple Sclerosis Introduction: MicroRNAs (miRNAs) are small and non-coding ribonucleic acids that play critical roles in regulation of host genome expression at post-transcriptional level. An individual miRNA is able to down-regulate multiple targeted mRNA transcripts. Therefore, minor changes in a miRNA expression may lead to significant alterations in the expression of different genes. During last two decades, miRNAs have emerged as key regulators of immune cell lineage differentiation, maturation, maintenance of immune homeostasis, and normal function. Multiples sclerosis (MS) is a chronic inflammatory disease that is characterized by infiltration of lymphocytes into the central nervous system (CNS), demyelination and axonal degeneration. Although causes of MS are still unknown, it is widely accepted that novel drug targets need to focus on both decreasing inflammation and promoting CNS repair. Recent researches about MS disease have shown that miRNAs are dysregulated in the immune system and CNS, which shows their role in the MS pathogenesis. Conclusion: Identification of specific expression patterns of miRNA in autoimmune diseases and a further comprehensive understanding of their role in the pathogenesis of different diseases offers promise of not only novel molecular diagnostic markers but also new gene therapy strategies for treating inflammatory autoimmune diseases. REVIEW_ARTICLE بیماری آلزایمر: اثر مسیر سیگنالینگ Nrf2 روی مرگ سلولی ناشی از استرس اکسیداتیو مقدمه: یک شیوع روز افزونی از بیماری آلزایمر وجود دارد. رسوب آمیلوئید بتا و سمیت عصبی یک نقش مؤثری را در بیماری آلزایمر ایفا می کنند. تصور می شود که استرس اکسیداتیو نقش اصلی را در بیماریی‏زایی دارد که منجر به تولید گونه‏های اکسیژن فعال و ایجاد آسیب‌هایی به ماکرومولکول‌ها در سلول‏های هدف می شود. گزارش شده است که فاکتور هسته‌ایی اریتروئید2 مرتبط با فاکتور 2 (Nrf2) یک تنظیم کننده‎ی کلیدی سیستم‌های دفاعی قابل القای داخلی در بدن است و سطح بسیاری از آنتی اکسیدان‏ها مانند گلوتاتیون S-ترانسفراز را افزایش می‌دهد. در شرایط آسیب اکسیداتیو، Nrf2 به هسته منتقل می‌شود و به عنصر پاسخ آنتی اکسیدان (ARE) متصل می شود و توالی را برای آغاز رونویسی از ژن‏های حفاظت کننده سلول افزایش می‌دهد. این مطالعه مروری بر مکانیسم‌های سلولی تنظیم Nrf2 تمرکز می‌کند و در مورد ارتباط بین تنظیم Nrf2 و بیماری آلزایمر بحث می‎نماید. نتیجه گیری: به طور کلی ما پیشنهاد می‌کنیم که فعال‌سازیNrf2-ARE یک مسیر محافظت‌کننده نورونی جدید است که می تواند به عنوان یک استراتژی درمانی امید بخشی برای درمان اختلالات تحلیل برندۀ عصبی مانند بیماری آلزایمر در نظر گرفته شود. http://shefayekhatam.ir/article-1-661-fa.pdf 2015-06-06 145 156 10.18869/acadpub.shefa.3.1.145 بیماری آلزایمر پپتیدهای آمیلوئید بتا استرس اکسیداتیو گونه‏های اکسیژن فعال Alzheimer’s Disease: The Effect of Nrf2 Signaling Pathway on Cell Death Caused by Oxidative Stress Introduction: There is an increasing prevalence of Alzheimer's disease (AD). Amyloid-beta deposition and neurotoxicity play an effective role in AD. Oxidative stress is thought to be central in the pathogenesis that leads to production of reactive oxygen species and causing damages of the macromolecules in target cells. It has been reported that the nuclear factor erythroid 2 related factor 2 (Nrf2) is a key regulator of endogenous inducible defense systems in the body and increase the level of many antioxidants, including glutathione-s-transferase. Under oxidative damage conditions, Nrf2 translocates to the nucleus, binds to the antioxidant response element (ARE), and enhances sequence to initiate transcription of cytoprotective genes. This review focuses on cellular mechanisms of Nrf2 regulation and discusses the relationship between Nrf2 regulation and AD. REVIEW_ARTICLE مروری اجمالی بر علل اختلال طیف اوتیسم مقدمه: امروزه بسیاری از محققان، اختلال طیف اوتیسم را برای توصیف اختلالات‌ رشدی فراگیر به کار می‌برند. در این مدل، تفاوت‌های سه اختلال رشدی فراگیر رایج پیشین شامل اختلال اوتیسم، اختلال آسپرگر و اختلالات رشدی فراگیر از جهات دیگر نامشخص، به سطوح متفاوت اختلال و نه نوع اختلال اشاره می‌کند. بر اساس اهمیت و پیش‌آگهی ضعیف و عدم آگاهی لازم در میان افراد جامعۀ ایرانی، این مقاله در مورد مهم‌ترین علل ایجاد اختلال طیف اوتیسم بحث می‌کند. نتیجه گیری: هنوز هیچ علت واحدی برای اختلال طیف اوتیسم شناخته نشده است. اگرچه اختلال طیف اوتیسم ممکن است در اثر کنش و واکنش عوامل عصبی زیستی و محیطی با استعداد ژنتیکی ایجاد شود. http://shefayekhatam.ir/article-1-662-fa.pdf 2015-06-09 157 166 10.18869/acadpub.shefa.3.1.157 اختلالات فراگیر رشد کودک وراثت نوروبیولوژی A Brief Review on the Causes of Autism Spectrum Disorder Introduction: Nowadays, many researchers apply autism spectrum disorder (ASD) to describe pervasive developmental disorders. In this model, the differences of previous three common pervasive developmental disorders, including autism disorder, Asperger’s disorder, and pervasive developmental disorder not otherwise specified, point to the various levels and not the type of disorder. According to the importance and poor prognosis as well as the lack of awareness of this disorder among Iranian people, this article discusses the most important causes of ASD. Conclusion: There is no single cause has still been known for ASD.
