اثر لیتیوم کلراید بر سطح سرمی فاکتور نوروتروفیک مشتق شده از مغز، فاکتور نکروزدهندۀ توموری آلفا و وزن مرطوب مغز در مدل حیوانی افسردگی
Effect of Lithium Chloride on Serum Levels of BDNF, TNF-α, and Wet Weight of Brain in an Animal Model of Depression

<p style="text-align: justify;"><strong>مقدمه:</strong> پاتوفیزیولوژی افسردگی یک موضوع چالش برانگیز است. ضایعات هیپوکامپ میتوانند باعث ایجاد افسردگی شوند و نیز تغییراتی را در سطوح چندین سایتوکین از جمله فاکتور نوروتروفیک مشتق شده از مغز (BDNF) و فاکتور نکروزدهندۀ توموری آلفا (TNF-α) ایجاد کنند. هدف از انجام مطالعۀ حاضر بررسی مکانیسم افسردگی القاء شده توسط مسمومیت با تری متیل تین (TMT) و مطالعۀ اثر تجویز مزمن لیتیوم کلراید (Li) بر روی افسردگی در این مدل حیوانی است. <strong>مواد و روشها:</strong> حیوانات به گروههای TMT+Li20، TMT+Li40، TMT+Li80 و TMT+ Saline تقسیم شدند که 8 میلی گرم/کیلوگرم TMT و به ترتیب 20، 40 و 80 میلی گرم/کیلوگرم لیتیوم کلراید یا سالین را به مدت 14 روز دریافت کردند. به منظور سنجش میزان افسردگی، مدت زمان بی حرکتی در آزمون شنای اجباری (FST) و آزمون معلق ماندن دم (TST) اندازهگیری شد. سپس سطح سرمی TNF-α،BDNF و وزن مرطوب مغز اندازهگیری شد. <strong>یافتهها:</strong> مدت زمان بیحرکتی در FST و TST در موشهای صحرایی که TMT دریافت کرده بودند طولانیتر بود، در حالیکه موشهای صحرایی دریافت کنندۀ لیتیوم کلراید بیحرکتی معنیدار کمتری را نسبت به گروه TMT+Saline نشان دادند. به علاوه تجویز لیتیوم کلراید سطح سرمی BDNF و وزن مرطوب مغزها را افزایش داد و سطح TNF-α را در مقایسه با گروه TMT+Saline کاهش داد. <strong>نتیجه گیری:</strong> کاهش BDNF یا افزایش فاکتورهای التهابی (به ویژه TNF-α) همراه با علایم افسردگی در مدل مسمومیت با تری متیل تین رخ داد. از طرف دیگر، تجویز مزمن Liممکن است سایتوکاینها را تعدیل کند و علایم رفتاری را بهبود بخشد.</p>
<p style="text-align: justify;"><strong>Introduction:</strong> Pathophysiology of depression is a controversial issue. Hippocampal lesions could lead into depression as well as to changing the levels of several cytokines, including brain-derived neurotrophic factor (BDNF) and tumor necrosis factor alpha (TNF- α). The present study was aimed to investigate the mechanism of depression induced by trimethyltin (TMT) intoxication and to study the effect chronic administration of lithium chloride (Li) on depression in this animal model. <strong>Materials and Methods:</strong> The animals were divided into TMT+Li20, TMT+Li40, TMT+ Li80 and TMT+Salin groups, which were received 8 mg/kg TMT and 20, 40 or 80 mg/kg of Li or saline, respectively, for fourteen days. In order to define the depression level, the immobility time of the rats was measured in the forced swim test (FST), and tail suspension test (TST). Then, the serum level of TNF-α, BDNF and wet weight of the brains were measured. <strong>Results:</strong> The immobility time in FST and TST was longer in the rats who received TMT, whereas the rats receiving Li showed a significantly less immobility compared to the TMT+Saline group. In addition, Li administration increased the serum level of BDNF and wet weight of the brains and decreased the TNF-α level compared to the TMT+Saline group. <strong>Conclusion:</strong> The decrease in BDNF or the increase in inflammation factors (especially TNF-α) occurred in accompany by depression symptoms in TMT intoxication model. On the other hand, chronic administration of Li may modulate the cytokines and amelioration of behavioral symptoms.</p>
