اثرات ضد التهابی نانو- کورکومین بر روی رده سلولی گلیوبلاستوم

مقدمه: گلیوبلاستوم مولتی فورم به‌عنوان یک تومور مغزی مهاجم شناخته می‌شود که با ویژگی میزان بالای عود مجدد مشخص می‌گردد. استراتژی‌های درمانی اخیر برای درمان گلیوبلستوما مولتی فورم موثر نیست، بنابراین گزینه‌های درمانی هدفمند نوین به شدت مورد نیاز می باشد. اخیرا، تحویل دارو بر اساس حامل‌های نانو به خاطر ویژگی‌هایی مثل توانایی عبور از سد مغزی نخاعی، آزادسازی پیوسته دارو، افزایش فعالیت زیستی و افزایش حلالیت داروها مورد توجه می‌باشند. به این منظور، ما اثربخشی یک شکل نانو از کورکومین در مقابل شکل آزاد آن را بر ترشح پروفایل سایتوکین‌های پیش التهابی IL6 و TNF-α در رده سلولی U87 بررسی کردیم. مواد و روش‌ها: لاین سلولی U87 از بانک ذخایر ژنتیک ایران خریداری و در محیط DMEM/F12 با 10 درصد سرم جنین گاوی و پنیسیلین/ استرپتومایسین 1 درصد کشت داده شد. برای سنتز نانوونیوزوم حاوی کورکومین، از روش هیبریداسیون لایه نازک استفاده شد. برای ارزیابی تولید IL6 و TNF-α به روش الایزا، سلول‌های U87 با 84.87 µg/ml نانو کورکومین و 47 µg/ml کورکومین آزاد تیمار شدند. یافته‌ها: نتایج ما نشان داد که تولید IL6 و TNF-α هنگام درمان با کورکومین نانو فرم و فرم آزاد به طور معنی‌داری کاهش یافت. جالب توجه است که ما مشاهده کردیم که نانو کورکومین می‌تواند به‌طور قابل توجهی ترشح IL-6 و TNF-α را در مقایسه با گروه کورکومین آزاد مهار کند. نتیجه‌گیری: واضح‌ترین یافته‌ای که از این مطالعه به دست می‌آید این است که نانو کورکومین اثرات ضد التهابی بر روی گلیوبلاستوم دارد.
